Not sure if possible to rent a car in Athens and drop it at Istanbul. In any case this would be pretty expensive. Check to drop it at Thessaloniki and take the train (or plane) to Istanbul. Now about road tips in Greece apart from Athens. You take the northern route, so you should visit Delphi and Vergina. Do not miss Vergina, at any cost, the tomb of Philip Ii, father of Alexander the Great. Unique place and museum. Spend fewer days in Athens and more in Thessaloniki while in Greece. Much friendlier place.

thanks for having an interest in visiting greece, we all appreciate it.

as for the car rental question:
im not sure that you can rent a car in greece, and to drop it on turkey. (due to the 2 different countries being with different laws+systems as a whole).

nonetheless, you should search for car rental companies that allow for a cross-country rental service, so that you may do that ''rent in greece, drop it in turkey'' business with your rental car.

as for sightseeing in greece, you can see many things in athens:

1) see the acropolis, and the temple of the parthenon on it.
also the temple of hephaestus or the ''thiseion'' is another great sight, and its a rare case of a fully preserved, intact ancient greek temple.

anothe fine sight is the ''tower of the winds'', which is located in the ''plaka'' area of the acropolis, within the ''roman market''.

2) the place of ''pnika'' or ''πνυκα'' is another fine natural sight: its the hill where ancient greek politicians used to speak from, and where they decreed many of their laws+topics.

3) you should visit the oracle of delphi, which is located in the area northwest of athens, close to amphissa. (but before it).
also, like exohoritis said above, you should also see the royal tombs of vergina, where phillip II, the father of alexander the great rests.
this is a great place of heritage for the ancient greek kingdom of macedonia.

4) if you wish to go to thessaloniki, you should see the epic building of the rhotonda (ροτόντα), and the white tower too, while the byzantine museum of thessaloniki is also a nice place to see as well.

as for turkey, it doesnt have many interesting things, apart from the greek orthodox temple of ''hagia sofia'', (αγια σοφια), which exists in constantinople (not istabul).

I would like to inform you that trafic in the city of Constantinople is Huge. My daughter visited the city 2 weeks ago and as she described , for a few minutes drive in normal cases , 5-6 km to get an idea , was taking them 2hours and 15 minutes to cover. Next day she used the metro and it was much much faster.Dont forget that Constantinople has over 15 million population.so DONT use a car there !!!

Think it is highly unlikely to be able to rent a car here and drop it in Turkey. Even if possible it would probably be very expensive. Try what was suggested. Rent car in Athens, drive to Thessaloniki and then catch a bus or plane to Istanbul. As to what to see, I would suggest Acropolis and the museum, Delphi, the Athens archaeological museum, Meteora if you have time, Vergina for sure and Thessaloniki with its Byzantine walls museum Rotonda etc.
Istanbul (Constantinople) has lots of lovely places too. Agia Sofia is a must together with a number of sites of interest by the Bosporus. Traffic is truly horrendous, so probably do not drive there.
